For most purposes, we actually do know most of the sequence. We are sequencing specific regions that mutate either very slowly (to establish phylogeny) or very easily (to do paternity tests), but the sequence around those regions is conserved across species/individuals. So we use those conserved regions as template for the primers. Alternatively, for applications like whole-genome sequencing, the strategy is to cut the genome into smaller DNA fragments and to integrate them into known DNA molecules. Then you can use the sequence of the known DNA molecule to design your primers and sequence each fragment separately.After that, you have to analyse the sequence of each fragment, looking for overlapping sequences. Fragments which have overlapping sequences were probably sitting next to each other in the genome. With enough overlapping fragments, the whole genome can be reconstructed. This analysis is done by computers

Primer is very useful in unpainted walls,it helps get an even color coverage,it is usually the same paint diluted with either water or turpentine (oil paint). Since the work is the same,I rather use regular paint and if it does'nt look too good ,I will give it a second coat.
